Lofepramine: a return to tricyclic antidepressives

Abstract

Lofepramine (Gamanil - E.Merck) is a new tricyclic antidepressive, a precursor of desipramine (desmethylimipramine). Its development aimed to retain the efficacy of standard tricyclics with a more rapid onset of action and fewer anticholinergic effects. Has this been achieved?
